ABSTRACT

Objective To pay more attention to the adverse drug reactions caused by tigecycline. Methods Four cases in our hospital which happened acute or subacute liver failure after the uses of tigecycline were analyzed, including age, basic disease, drug combination,the time when liver function failure happen ,dosage of tigecycline and the course of treatment. Results The aged of four patients was 49-77 years old, the average age (67.5±12.79)years; the time when liver failure happened from 6 d to 20 d, the average(11.75±6.24) d; the course of treatment was 9 d to 23 d, the average(9.75±6.63) d; the total dose of tigecycline was 900 mg to 3650 mg, the average (1812.5±1243.23) mg.The four patients have many basic diseases, combined with many drugs, all of them have mechanical ventilation. Conclusion More attention should be paid to adverse events caused by tigecycline.

ABSTRACT

Objective To investigate the therapeutic effect of microencapsulated xenogenic hepatocytes transplantation on rat fulminant hepatic failure(FHF).Methods Isolated guinea pig hepatocytes were microencapsulated with alginate-chitosan-polyethyleneglycol(ACP).The microencapsulated hepatocytes(MCHC) were implanted into the peritoneal cavity of Wister rats 24 hrs before the FHF model,which was surgically induced by 90% hepatectomy.After 24hrs of the FHF,the serum ALT,total bitirubin(TB),blood suger(BG) and PT were determined,and the microscopic appearance,function and nember of the implanted microencapsulated hepatocytes were observed,as well as the survival time and life quality of the FHF rats. Results Compared with the control group,the rats in the MCHC implantation group showed milder symptoms of central nervous system and less serious ascites.The ALT,BS, TB,and PT in the MCHC implantation group were all improved significantly (P

ABSTRACT

Objective To establish a new model of acute hepatic failure in canines for bioartificial liver treatment research . Methods Acute hepatic failure was induced by end to side portocaval shunt combined with common bile duct ligation and transection. The survival rate, liver function, ammonia, Fischer's ratio (BCAA/AAA) and pathological change of injury liver were investigated. Results The ammonia, ALT, AKP, total bilirubin and PT were elevated dramatically and Fischer's ratio declined significantly 14 days after the operation. The survival rate of the models decreased with time 14 days after the operation. Pathological examination of liver tissues revealed evidence of cholestasis, inflammation with accumulation of neutrophils, degeneration and necrosis of hepatocytes.Conclusions End to side portocaval shunt combined with common bile duct ligation and transection can produce a satisfactory acute hepatic failure model in canines, which could be used in the study of bioartificial liver.
